Serotyping of E. coli Strains

We perform O and H typing for classifying E. coli for pathogenicity

O serotyping

Performed using antisera generated against E. coli serogroups designated O1-O187 with the exceptions of O31, O47, O67, O72, O94, O122 as these are not designated.

Performed following the procedure published by Orskov et al. 1977. Serology, Chemistry, and Genetics of O and K antigens of Escherichia coli. Bacteriol. Rev. 41:667-710. The standard strains used are described in the paper.

H typing

Performed by PCR-RFLP analysis (Polymerase Chain Reaction- Restriction Fragment Length Polymorphism) of fliC gene responsible for flagella

Performed by the method described by Machado et al. 2000. Identification of Escherichia coli flagellar types by restriction of the amplified fliC gene. Res. Microbiol. 151:535-546

Detection of genes to determine O types and H types

  • O157, O145, O121, O113, O111, O103, O45, O26 and many others
  • H7
